The researchers extracted a lectin from the seeds that has strong HIV-1 reverse transcriptase inhibitory activity. This discovery also provides data to support the application of H. mutabilis. We extract lectins from H. mutabilis seeds. We optimize the reported extraction method to avoid the problems of product impurity, long experimental period, etc., encountered in traditional lectin production. We then use a combination of gel filtration chromatography and ion exchange chromatography to further purify the crude extract from the seeds. After the purity and content test, the optimized production method improves the yield and purity of lectin.
Upon the client's request, we test the physicochemical properties of the extracted lectin. We detect its molecular weight by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The lectin extracted from H. mutabilis seeds is found to have binding specificity for galacturonic acid. We test the inhibitory effect of metal ions and different kinds of carbohydrates on their agglutination activity. In addition to this, agglutination activity and others are also tested.

Results: In this study, a lectin was extracted from dried H. mutabilis seeds. It was purified using a combination of gel filtration chromatography and ion exchange chromatography. It was found to have a molecular weight of 150-kDa by SDS-PAGE. the results of sugar inhibition experiments showed that the lectin had binding specificity for galacturonic acid. It also exhibited strong HIV-1 reverse transcriptase inhibitory activity and weak antiproliferative activity against tumor cells. However, it showed no inhibitory activity against Fusarium oxysporum, Mycosphaerella arachidicola, Valsa mali, etc.
