Natamycin, also known as natamycin, is a naturally occurring antifungal agent with antibiotic properties produced by Actinomyces spp. This compound exhibits broad-spectrum activity against a wide range of fungi and yeasts. Although natamycin has significant antifungal activity, in practical application, an in-depth study of its antifungal mechanism of action and symptomatic use is essential. This is because different fungi have different sensitivities to natamycin and may be resistant to it. Results: This article focuses on an immunoassay method for the detection of antifungal drug natamycin traces in food. The method employs specific antibodies that selectively bind to natamycin and quantifies its concentration in the sample by measuring the signal generated from the immunoreaction, thereby enabling rapid screening. The article verifies that cyclodextrins (CDs) as food additives do not have a significant effect on the determination of natamycin. The presence of CDs in the natamycin assay does not alter the natamycin measurement. In addition, since organic solvents destroyed the CD-natamycin complexes, the CD-natamycin complexes were destroyed during the extraction of natamycin from food samples and did not affect the detection of natamycin. The article also developed an ELISA variant for the extraction of natamycin from different food matrices. The extraction process was optimized, solvents were used, and different food matrices were tested. The recovery of the extraction process was 72-106% with a variability of less than 12%. The ELISA variant had the highest sensitivity (IC50 = 0.12 ng/mL) with a detection limit of 0.02 ng/mL for the natamycin assay. Thus, this ELISA variant can be used to extract natamycin from different food matrices with high sensitivity and detection limits. The recovery and variability of the extraction process were also within acceptable limits.
