Gangliosides are acidic glycosphingolipids that form lipid rafts in the outer layer of the plasma membrane, especially in neuronal cells of the central nervous system. GT1 induces the degeneration of dopaminergic neurons, which may contribute to the development or progression of Parkinson's disease. GT1 inhibits antigen- or mitogen-induced T-cell proliferative responses and has been identified as a receptor for botulinum toxin, a rare toxin with severe physiological consequences. The ganglioside is a free radical scavenger that protects against brain mitochondrial DNA damage, seizures, and lipid peroxidation caused by reactive oxygen species generators.

CD BioGlyco provides high-quality Trisialoganglioside (GT) Analysis Services. We offer quadrupole time-of-flight mass spectrometry (QTOF MS) for the analysis of GT1 in negative ion mode. Gangliosides are characterized by nanoESI MS screening and tandem MS (MS/MS), and structure is characterized using collision-induced dissociation (CID) sequencing.
Use tissue of the same quality as the starting material for ganglioside extraction. Extraction and purification of samples are carried out under the same conditions. Tissues are homogenized in ice-cold water before the extraction of gangliosides.
Crude gangliosides isolated and purified from tissues are analyzed by HPTLC and laser densitometry. Quantification of total ganglioside concentrations is performed spectrophotometrically.
For QTOF MS analysis, stock solutions of individual ganglioside extract at certain concentrations are prepared by dissolving the dried material in pure methanol. Treated supernatants are collected and subjected to nano-ESI QTOF MS and MS/MS analysis at low energy by collision-induced dissociation (CID).
All mass spectra are recorded in negative ion mode, previously shown to be the best choice for this class of molecules, as gangliosides are readily deprotonated. All parameters are adjusted to optimal ranges for efficient ionization and minimized source fragmentation.
