- Character observation: The sample should be a colorless or almost colorless clear liquid.
- Sample identification:
- We add an alkaline copper tartrate test solution to the sample and then observe whether there is red precipitation.
- We observe whether the retention time of the main peak in the chromatogram is consistent with that of the control by high-performance liquid chromatography (HPLC).
- pH value: The pH value of the sample should be 3.5~5.5.
- Guanine and other related substances: The content of guanine and impurities in the sample is detected by HPLC.
- 5-Hydroxymethylfurfural: The content of 5-hydroxymethylfurfural in the sample is detected by HPLC. The amount of 5-hydroxymethylfurfural should not be more than 0.03% of the labeled amount of glucose.
- Determination of content:
- Aciclovir: We detect the content of aciclovir contained in the sample by HPLC.
- Glucose: We detect the optical rotation of the sample and calculate the amount of glucose in the sample.
- Other tests: Microbiological tests, bacterial endotoxins, heavy metals, molar concentration of osmolality, etc.

Results: In the present study, the researchers analyzed the stability of parenteral acyclovir when diluted in dextrose as a high-volume intravenous liquid formulation by HPLC and other methods. The results showed that when acyclovir was diluted in glucose solution, the level of drug loss ranged from 0.27% to 1.03% of its initial content. This study demonstrates the suitability of HPLC for the correlation analysis of acyclovir.
