Lectins are highly abundant in C. ensiformis seeds. We mainly extract lectins from seed species. Using C. ensiformis seeds as raw material, we summarize the published extraction process and optimize it. The optimized extraction method greatly shortens the experimental period.
Most lectins belong to proteins, so many protein purification methods are used to purify lectins. There are many methods for lectin isolation and purification, mainly including ion-exchange chromatography, Gel-filtration Chromatography, affinity chromatography, and other methods. We purify C. ensiformis lectin by Ion Exchange Chromatography and Affinity Chromatography. C. ensiformis lectin is heat stable. We compare the hemagglutination activity of lectins obtained by extraction at different temperatures to determine the optimal temperature for purification. The optimized method greatly improves efficiency.
After purification, we study the physicochemical properties of C. ensiformis lectin with high-performance liquid chromatography, gas chromatography-mass spectrometry, and other instruments.

Results: In the present study, Con A was extracted and purified from the seeds of C. ensiformis. The crude extract was obtained using a sodium chloride solution. The crude extract was obtained using sodium chloride solution, followed by affinity chromatography to purify Con A. Its purity was determined by gel electrophoresis (SDS-PAGE). The results showed that ConA displayed three bands: α chain (25.5 kDa), β chain (14 kDa), and γ chain (12 kDa).
