  • CAL production service

The seeds of leguminous plants contain more lectins. We mainly use C. arietinum seeds as materials to extract lectins. The crude extract of lectin is obtained by Tris-HCl extraction. Afterward, purification is carried out by precipitation and dialysis. Ion Exchange Chromatography has a very high resolution, high recovery, and a wide range of applications. We use ion exchange chromatography for further purification of lectins.

The molecular weight of C.arietinum lectins varies from species to species. The physicochemical properties of the extracted lectins will be tested according to client requirements. We have advanced types of equipment such as amino acid auto analyzer, Fourier-transform infrared spectroscopy, etc. Amino acid composition, sugar-binding specificity analysis, secondary structure detection, molecular weight detection, etc are all accomplished.

Results: In the present study, C. arietinum seed powder was used as raw material for the extraction of lectins. Tris-HCl extraction was followed by dialysis. This was followed by further purification by sequential ion exchange chromatography including diethylaminoethyl (DEAE)-cellulose and Sulphopropyl (SP)-Sephadex matrix. The lectin was finally successfully extracted. The molecular weight of the lectin subunit was determined to be 35 kDa. The molecular weight of the lectin in the seeds of different varieties of C. arietinum may vary.
