The capsular polysaccharide (CPS), which gives enhanced resistance to host defenses such as complement-mediated death and phagocytosis, is a crucial virulence component during invasive infections. Furthermore, the similarity of certain CPS to host molecules enables bacteria to employ a molecular mimicry technique, which further supports survival. Sometimes the capsule is made of polymers that resemble glycosaminoglycans (GAGs). GAGs are long unbranched polysaccharides found in vertebrates that are typically present on the surface of cells or in the extracellular matrix, where they play an important role in cell growth, adhesion, differentiation, and proliferation. GAGs, including chondroitin sulfate (CS), heparin, and hyaluronic acid, are economically lucrative biopolymers with a variety of uses in the medical, veterinary, pharmaceutical, and cosmetic industries. Chondroitin-like polysaccharides were found to be suitable substrates for GlcA- and GalNAc-transferase activities in E. coli K4, indicating their potential involvement in capsule polysaccharide synthesis.

CPS production has traditionally relied on isolating it from animal tissues, which can be challenging due to the risks of viral and prion contamination and the associated high costs. In 1996, the first study on cultivating E. coli K4 and exploring the structure of its cell-bound and cell-free K4 polysaccharide was published. However, there is currently limited research on the physiological aspects of E. coli that connect dietary requirements and growth conditions to CPS production.
