It is an essential idea of 1H,1H correlated spectroscopy (or 1H,1H-COSY) to connect protons that are scalarly (or through-bonds) coupled and have a coupling constant value (J) different than zero. With the exception of some precise geometric conditions, which is seldom the case in carbohydrates, these protons are normally separated by two (2J) or three (3J) chemical bonds, whereas protons separated by four linkages (or more) are often not connected. Like other homonuclear spectra, the COSY spectrum is symmetrical around the diagonal, therefore its densities may be separated into those that form the diagonal and those that are off the diagonal.

A useful method for identifying signals that are generated by adjacent protons often up to four bonds is 1H,1H-COSY. Protons show correlations when spin-spin interaction is present. However, where there is no connectivity, no correlation is expected. The most popular type is DQF-COSY, which has gradient-enhanced double-quantum coherence.
