The preferred methods for analyzing stable isotope labeling patterns in cell metabolites are NMR and MS. The biosynthetic network may be quantitatively described by the measurement of isotope enrichment in metabolites, which also permits flow analysis in reaction to outside disruptions. Total correlation spectroscopy (TOCSY), like COSY, is a homonuclear 2D analysis. Contrary to COSY, the identification of related spins is not limited to immediate neighbors. By altering the isotropic mixing scenario during spin-locking, the TOCSY experiment creates cross-peaks between all hydrogen atoms that are part of a continuous chain of linked spins.

The more well-known COSY experiment is usefully supplemented by the 2D TOCSY experiment. TOCSY can offer correlations between all of the protons that make up a spin system, whereas COSY often develops correlations between protons spanning two to three chemical bonds. Unlike COSY, which only establishes correlations between geminal or vicinal protons, TOCSY establishes correlations between all protons in a particular spin system. As long as there exist couplings between each intervening proton, correlations between far-off protons can be observed. This is very helpful for locating protons on amino acids or sugar rings: The connection between every proton on a given sugar ring and every other proton on the same ring will always be the same, but not for protons on different rings.
