Structure and Function of Human Serum Albumin (HSA)

HSA consists of a single chain of 585 amino acids. Its secondary structure is highly flexible, with 67% α-helices and 17 disulfide bridges with 6 turns, acting as cross-linkers for the three homology domains. It acts as a free radical scavenger under inflammatory conditions and is involved in processes such as blood clotting and wound healing. Furthermore, it has antioxidant properties and heparin-like activity by enhancing antithrombin IIl activity. The albumin molecule has many charged amino acid residues. Therefore, sodium ions and other cations are strongly attracted to albumin.

HSA Conjugation Services at CD BioGlyco

  • Acquisition of polysaccharides and HSA

According to the needs of the experiment, suitable polysaccharides and HSA are obtained. HSA is filtered through deionized water at room temperature, sterile filtered, and freeze-dried before experiments.

  • Polysaccharide and HSA conjugation

Molecular weights of HSA and polysaccharides are determined by matrix-assisted laser desorption ionization time-of-flight mass spectrometry. The polysaccharide is activated with cyanogen bromide and derivatized with adipic dihydrazide, and conjugated to HSA by carbodiimide condensation. The reaction mixture is allowed to contain equal amounts of HSA and adipic hydrazide derivatives of polysaccharides. 1-ethyl-3-dimethylaminopropylcarbodiimide is added and the reaction mixture is kept slightly acidic. The mixture is dialyzed overnight and loaded onto the column. The composition of the polysaccharide-HSA conjugate is determined by chemical analysis. Fractions appearing in the column void volume are pooled and used to vaccinate mice.

  • Immunization and enzyme-linked immunosorbent assay (ELISA)

Female mice are subcutaneously injected into groups. Bleeding is performed after the number of injections is reached. Mice injected with saline are used as a control group. Then ELISA assay is performed.
