Cytosine is a pyrimidine heterocycle, which is one of the pyrimidine bases constituting nucleic acids. It is an important intermediate in pesticides, pharmaceuticals as well and fine chemicals. However, it has certain defects, such as poor lipid solubility, low bioavailability, certain toxic side effects, etc. Therefore, the development of new efficient and low-toxicity cytosine derivatives is of great significance. Some of the derivatives synthesized so far have produced good application value and social benefits. For example, in the field of biopesticides, cytosine derivatives have obvious inhibitory effects on diseases, insects, and grasses that affect the growth of crops. In the field of medicine, Other antiviral and anticancer cytosine derivative drugs have been synthesized using cytosine as an intermediate.

Cytosine derivatives are produced by various methods. Our one method is to convert uracil to 2,6-dichloropyrimidine, which is then decomposed into a mixture of chloropyrimidineamines. These chloropyrimidinamines are converted to a mixture of methoxypyrimidinamines by sodium methanol treatment. The isolated methoxypyrimidinamines are then acid hydrolyzed to cytosine to achieve separation of the desired components. Another route is the conversion of 2-thiouracil to dithiouracil followed by ammonolysis to 2-thiocytosine. Then it is heated with aqueous chloroacetic acid or in DMSO containing a small amount of sulfuric acid to produce cytosine and its derivatives. We modify cytosine in different directions such as C=C double bond, N-aromatization, selection of different catalysts, 1-position modification, etc. We also produce cytosine derivatives with new structures based on the produced derivatives. We provide the corresponding structural analysis data, including mass spectrometry (MS), NMR analysis, and high-performance liquid chromatography (HPLC), in addition to the products.
Currently, there are some shortcomings in the production of many cytosine derivatives, such as cumbersome routes, high cost of raw materials, low yields, etc. We also improve the production routes of different derivatives according to clients' requirements. We maximize the purity of the products and look for production routes with simple and easy-to-control operating, mild reaction conditions, and lower production costs.
