P. aeruginosa is a gram-negative, aerobic, non-spore-forming rod that can infect immune-competent and immune-compromised hosts, including people, with various diseases. Most bacteria in nature can cling to various surfaces and create biofilms. P. aeruginosa is a well-known producer of biofilms, and it contains several exopolysaccharides that are unique to biofilms, such as cellulose, alginate, Pel polysaccharide, and poly-N-acetylglucosamine (PNAG). Pel is a partly deacetylated cationic polysaccharide polymer comprising N-acetyl-d-glucosamine and N-acetyl-d-galactosamine. In nonmucoid strains, Pel is a crucial matrix component of the biofilm that is involved in both the start of surface attachment and the preservation of the biofilm's integrity.

Pel, which comes in two forms—a high-molecular-weight cell-associated form and a low-molecular-weight secreted form—is a positively charged, partly deacetylated, -1,4 linked N-acetylgalactosamine (GalNAc) polymer made mostly of GalNAc-GalN repeat units. CD BioGlyco has the ability to purify various molecular weights to offer custom P. aeruginosa Pel polysaccharide production services.
The protein byproducts of the pelABCDEFG operon are necessary for the production of pel-dependent biofilms. Although a clear connection between Pel production and immunological compromise and/or virulence has not yet been demonstrated, it has been demonstrated that the secretion of this polysaccharide increases the in vitro resistance of P. aeruginosa to aminoglycoside substances. Different P. aeruginosa strains have varying abilities to create biofilms. This is an excellent tool to look at possible antibacterial uses, CD BioGlyco offers a Polysaccharide Vaccine Development Platform to support our clients' glycan-related research.
