NADP is a coenzyme bound to a phosphate molecule in an ester bond, widely found in living organisms. NADP and its reduced form NADPH regulate reductive metabolism in a subcellular segregated manner and are central cofactors in lipid and redox homeostasis. NADPH is expressed in the reduced form through several enzymes in the cell and is recycled in the reduced NADPH and oxidized states NADP. NADP production in mitochondria is dependent on the phosphorylation of NAD (H) by NAD kinase 2 (NADK2), which is used to power biosynthetic pathways (including lipid synthesis) and redox homeostasis. NADK2 is important in the regulation of cellular redox state, energy metabolism, mitochondrial function, gene expression, and signaling pathways. NADK2-mediated mitochondrial NADP production is required for glutamate reduction and proline biosynthesis for cytoplasmic protein synthesis.
