Guanine is one of the four major nucleobases found in the nucleic acids DNA and RNA. It plays a key role in cells. Many guanine derivatives have been used as anticancer and antiviral agents for decades. Drugs made from guanine derivatives as pharmaceutical intermediates have shown particular efficacy against cancer, AIDS, etc. Among many guanine derivatives, some N2-substituted guanines also exhibit intriguing biological activities. Guanine analogs have been widely used in anticancer research for decades. They have a wide range of antitumor activities and act mainly by inducing apoptosis and inhibiting DNA synthesis. Several drugs with antitumor activity have been developed, such as 8-bromo-9-(β-D-xylofuranosyl) guanine, 8-hydroxyguanine, 7-(Butyn-2-yl)-7,8-dihydro-8-oxo-9-(β-D-xylofuranosyl) guanine, etc. We produce low-toxicity guanine derivatives that are used in cancer therapy research through various chemical reactions such as acylation and alkylation reactions.
