Guanine is one of the four major nucleobases found in the nucleic acids DNA and RNA. It plays a key role in cells. In addition to antitumor, it has been found that some N2-substituted guanine derivatives are better in other activities, such as thymidine kinase inhibitory activity, anti-microbial activity, and cell cycle protein-dependent kinase inhibitory activity. In addition, some N2-substituted guanines can also be used as synthetic intermediates for the preparation of molecular biology tools, diagnostic agents, etc. Based on the reported routes, some optimizations and improvements are made to produce N2-substituted guanine derivatives, such as various N2-alkylated guanines.
