2'-O-methyl RNA bases modification is a method of chemical modification specifically targeted towards oligonucleotides. In tRNA and other RNAs, 2'-O-methyl RNA is a naturally present modification, which can be part of custom-designed oligonucleotide configurations, where a 2'-O-methyl group is added to the ribose sugar of the oligonucleotide (Fig.1), altering its chemical structure and thereby changing its properties and functions. To include 2'-O-methyl modifications in an RNA sequence, simply place a lowercase "m" in front of the base; for example, mA, mG, mC, mU.
Compared to unmodified oligonucleotides, 2'-O-methyl modification is used to enhance the stability of oligonucleotides, prevent degradation by nucleases, and improve biological activity within cells. It also reduces nonspecific protein binding and increases the affinity and specificity of oligonucleotides for target RNA sequences. 2'-O-methyl modification finds widespread applications in various biomedical research and application fields, providing a useful tool for regulating the activity, selectivity, and stability of RNA.
