Oligonucleotide-based Affinity-plus Base Modification Service
What Is an Affinity-plus Base Modification?
Modified bases establish an epigenetic information layer that regulates gene expression. The modifications improve the regulatory and protective functions of DNA and don't affect the coding and base-pairing functions. Affinity Plus modified bases in a way that means locked nucleic acids (LNAs). It is a modified RNA monomer that implies a methylene bridge linking the 4' carbon and 2' oxygen of the RNA pentose ring (Figure 1). This bridge fixes the pentose ring in the 3' endo conformation. Add monomers of locked nucleic acid help to increase sequence melting temperature, and these bases also affect in vitro and in vivo nuclease resistance.

Affinity-plus Base Modification Services at CD BioGlyco
CD BioGlyco provides multiple types of Oligonucleotide-based Bases Modification Services, including Affinity-plus Base, Inosine, 2'-O-methoxy-ethyl Bases (2'-MOE), 2'-O-methyl RNA Bases, Fluoro Bases, 2-Aminopurine, 5-Bromo Du, etc.
- In affinity-plus base modification service, CD BioGlyco designs the sequence including 1−20 locked nucleic acid nucleotides to suit the client's study needs and offers LNAs services containing single- and double-stranded sequences at the same time.
- CD BioGlyco provides customized and predesigned DNA or RNA oligonucleotides and affinity plus qPCR probes. Products include primers, qPCR probes, plate-mounted high-throughput DNA, and next-generation sequencing adapters, with various specifications, which are produced in strict accordance with ISO 9001 and ISO 13485 quality systems for study.
- CD BioGlyco also provides highly efficient LNA synthesis services through two different approaches. The one is a linear strategy in that synthetic materials are usually selected for uridines (or any easily obtained RNA nucleoside). Another strategy is convergent, a sugar intermediate is synthesized as a glycosyl donor to couple with nucleobases in the process.

Applications
- Gene editing: Modified LNA can be used in single-base gene editing at the intended position, which can avoid mismatch repair DNA.
- Genotyping assays: LNA has been applied in diagnostic tools due to its high affinity for mismatch distinguish, which is introduced in multiplex SNP genotyping assays.
- Therapeutics: A variety of LNA oligonucleotides is assessed for their toxicity and pharmacokinetics. It is displayed that using LNA-based oligonucleotides therapeutically displays a preferential safety profile in the biotechnology field.
- Improve hybridization application: LNA is incorporated in probes, which can increase hybridization speed in both RNA and DNA.
