The Basic Roles of HEX Labeling
HEX, a popular orange dye, is commonly used as a probe for routine PCR as well as for quantitative and multiplex RT-PCR, for measuring changes in fluorescence, or for other applications in which HEX-odn interacts with biopolymers. In particular, HEX labeling is used for so-called multicolor combinatorial probe coding. HEX defines one of 13-15 possible DNA targets in a single PCR, the desired number of probes are labeled with one or 2-4 different fluorophores. Oligodeoxyribonucleotides (ODNs) containing Fluorophores are widely used in molecular biology and medical diagnostics, they are used as probes in real-time PCR (RT-PCR) DNA analysis.

HEX Labeling Service at CD BioGlyco

Any post-ammonolysis mixture of HEX-labeled oligodeoxyribonucleotides contained more than 25% hexachloroaryl acridine derivatives. The acridine by-product is not easily detectable or removable, and it may significantly distort the results of quantitative fluorescence analyses using HEX probes.
- We use a new, simple, and reliable post-synthesis deblocking method for HEX-labeled oligonucleotides. Designed to enable high yields of these fluorescent probes and to guarantee the absence of acridine by-products. Under PCR-like analog conditions, we significantly converted the hexachloroaryl acridine derivatives into non-fluorescent derivatives. Each step of the process was strictly controlled. - The HEX-labeled oligonucleotides were observed using the Scanning Tunneling Microscopy (STM) constant current mode, and then the samples were investigated using scanning tunneling spectroscopy (STS) to evaluate the performance, to ensure the best performance product is provided to our customers. Post-synthetic HPLC purification of HEX-labeled oligonucleotides is essential, where sensitivity or fluorescent signal differentiation (e.g., in quantitative and multiplexed assays) are critical.
