The structure of GT3 consists of a ceramide backbone linked to an oligosaccharide unit consisting of five sugar molecules. Taking human lung tissue samples as an example, our general analysis steps are as follows.
First, homogenize the lung tissue in a homogenizer. The lipid extract is then divided into two equal aliquots and perform a chromatograph. The column is eluted with chloroform, chloroform/methanol/water, chloroform/methanol/water, and chloroform/methanol/water, respectively. The last fraction contains the complex gangliosides, which are separated by anion exchange chromatography into monosialogangliosides, disialogangliosides, and GT. Individual gangliosides of the GT fraction are separated by thin-layer chromatography.
Acidic glycosphingolipids are analyzed by MALDI-QIT-TOF mass spectrometer in negative ion mode. Samples are prepared by mixing acidic glycosphingolipid samples directly on the target with a 2'-4'-6' trihydroxy acetophenone monohydrate matrix solution containing a diammonium hydrogen citrate hydrate mixture.
Sialic acid is determined using the resorcinol assay.
