Introduction

Human Milk Oligosaccharide (HMO), an unconjugated glycoprotein, is essential for the infant's protection provided by breast milk. An HMO is composed of glucose, galactose (Gal), N-acetylglucosamine, fucose (Fuc), and N-acetylneuraminic acid, which are the five monosaccharides that make up an HMO.

Fucosylated HMOs

Fucose and sialic acid commonly modify the backbones of HMO and N-glycans to produce a variety of recognition motifs, including blood-group antigens and Lewis antigens. Fucosyltransferase 2 (FUT2) is one of the enzymes that helps the mammary glands manufacture HMGs by catalyzing the transfer of a fucose residue through a 1,2-linkage to the glycans present in human milk.

The basic oligosaccharides found in breast milk are trisaccharides made of lactose to which fucose or sialic acid molecules have been joined by various glycosidic bonds. Consequently, two of the four oligosaccharides that are produced as a result of fucose being added to lactose by the enzymes 1,2 fucosyltransferase and 1,3/4 fucosyltransferase are each distinct. Fuc, for instance, generates 2'-fucosyllactose (2'-FL) when it is linked to Galactose Gal via a 1,2 glycosidic linkage, but not when it is linked to Gal via a 1,3 glycosidic linkage (3-FL).
