NMP consists of pentose ribose, base, and a phosphate functional group. According to the different bases, NMPs can be divided into adenine/guanine/cytosine/thymine/uracil monophosphates (AMP, GMP, CMP, TMP, UMP), which are the components of DNA or RNA. There are many non-nucleic acid nucleotides. For example, cyclic adenosine monophosphate (cAMP) is an important second messenger in the human body. In addition, guanosine tetraphosphate (ppGpp) and guanosine pentaphosphate (pppGpp) are related to the regulation of gene expression.
The enzymatic hydrolysis strategy is to split DNA or RNA into basic units by hydrolase to obtain the corresponding nucleotides. At present, 40% of nucleotides are derived from enzymatic hydrolysis of RNA.
Chemical synthesis has the advantages of a few steps, large production scale, short cycle, and high efficiency. Modified nucleotides with different biofunctions can be synthesized by chemical methods.
Bacillus subtilis, Corynebacterium ammoniagenes, etc. are mostly used in producing nucleotides through biological fermentation. At present, the industrial production of inosine monophosphate (IMP) and GMP have been realized.
